Filamin in the Z-disc
"Muscles are essential for locomotion and force-related mechanisms in organisms. The sarcomere is the basic unit of contractility in the muscle cell and allows for such forces. It is composed of antiparallel actin thin filaments and myosin thick filaments, with the thin filaments anchored at the Z-disc, a multiprotein complex, at both ends of the sarcomere. Many of the proteins present in the Z-disc are involved in myopathies when mutated. Filamin C is an actin filament crosslinker that is a core component of the Z-disc, and is mutated in muscle related diseases. Understanding its binding partners and dynamics is crucial to uncovering the pathology of these filamin-related myopathies. Using different filamin (cheerio) deletion mutants in the model organism Drosophila melanogaster, we identify unique indirect flight muscle phenotypes. These perturbations have consequences on the mutants' ability to use their flight muscles properly, as demonstrated by flight assay performance. Using biochemical, behavioural, and imaging techniques, we demonstrate the importance of filamin function in Z-disc stability and myofiber structural integrity"--

This leadership playbook has been published to coincide with the global release of the first PISA test results on creative thinking, the Creativity Collaboratives under way in England and the growing importance of creative thinking in countries across the world. Itoffers a framework for improving creative thinking based on the widely used five creative habits framework developed by the Centre for Real-World Learning. Near the end of his life, Roger Williams, Rhode Island founder and father of American religious freedom, scrawled an encrypted essay in the margins of a colonial-era book. For more than 300 years those shorthand notes remained indecipherable... ...until a team of Brown University undergraduates led by Lucas Mason-Brown cracked Williams' code after the marginalia languished for over a century in the archives of the John Carter Brown Library. At the time of Williams' writing, a trans-Atlantic debate on infant versus believer's baptism had taken shape that included London Baptist minister John Norcott and the famous Puritan "Apostle to the Indians," John Eliot. Amazingly, Williams' code contained a previously undiscovered essay, which was a point-by-point refutation of Eliot's book supporting infant baptism. History professors Linford D. Fisher and J. Stanley Lemons immediately recognized the importance of what turned out to be theologian Roger Williams' final treatise.
